Title: HiPerDART: Development of High Performance Diagnostic Array Replication Technology for prognostic prediction in colorectal cancer (PS09/01037)

Summary: The HiPerDART project aims to develop, using a new printing technology called Supramolecular NanoStamping (SUNS), a higher level of microarray technology that promises to dramatically improve the reliability of medical evidence. This technology will be tested in a clinical application to generate a prognostic and predictive tool based on a broad set of genomic markers, selected using novel techniques of bioinformatics.
Patients diagnosed with stage II colorectal cancer may benefit from a molecular profile that would indicate the need for adjuvant chemotherapy. Inherent in this approach is the assumption that each tumor and the mucosa where is originated, contain signatures genetic and epigenetic information that, at the time of diagnosis, can address the biological behavior of tumor gene expression, the abundance of MicroRNAs, the genetic alterations in DNA (SNPs / CNVs / LOH) and DNA methylation will provide a combination of more sensitive and specific biomarkers to identify those patients with more aggressive tumors who may benefit from adjuvant chemotherapy. HiPerDART Project will use available data on expression and SNPs / CNVs to develop predictors.
We intend to expand the domain of genetic information in DNA methylation and microRNA profiles to improve the accuracy of prediction. The profile obtained will be implemented on a SUN microarray and validated in a large independent sample of colorectal tumors. Biology Systems tools as well as modeling of networks will be used to understand the relationships between genetic and epigenetic expression and its regulators (DNA methylation and microRNAs).
